Short Communication Effects of Histamine on Phosphoinositide Metabolism in Chick Cerebral Cortex
نویسندگان
چکیده
Effects of histamine (HA) and agonists of HA receptors on phosphoinositide metabolism in chick cerebral cortex have been studied using two approaches – measurement of inositol 1,4,5-trisphosphate (IP ) level by a specific and sensitive IP receptor radioassay, and analysis of [ H]inositol phosphates accumulation in cortical slices prelabeled with myo-[ H]inositol. HA concentration-dependently elevated IP levels in slices of chick cerebral cortex. The effect of HA was mimicked by 2-methylHA, a selective agonist of H -HA receptors, and blocked by mepyramine, an H receptor antagonist. 4-MethylHA and R -methylHA, selective agonists of H and H -HA receptors, respectively, did not affect IP level in the chick cerebrum. In cerebral cortical slices prelabeled with myo-[ H]inositol, 2-methylHA significantly stimulated [ H]inositol phosphates accumulation, whereas HA only slightly and non-significantly increased phosphoinositide metabolism. It is suggested that phospholipase C-coupled H -HA receptors are present in the chick cerebral cortex, yet their number seems to be a small one.

متن کاملEffect of acute exposure to ethanol on distribution of NR1 subunit of NMDA receptor of glutamate in cerebral cortex of chick embryo
Introduction: There is considerable evidence that glutamate-mediated excitatory neurotransmission plays an important role in mediating the behavioral actions of acutely administered ethanol. The aim of the present study was to investigate the effect of acute ethanol exposure on NR1 subunit of NMDA (n-methyl-d-aspartate) receptor distribution in the cerebral cortex of chick embryo on the 10th...
متن کاملPreliminary Communication Desensitization of H2-like Histamine Receptors Stimulating Cyclic Amp Formation in the Chick Cerebral Cortex
Histamine potently stimulates cyclic AMP formation in slices of the chick cerebral cortex. Pretreatment of the tissue slices with 10 M histamine for 2–30 min led to a time-dependent attenuation (when compared with values observed in the control tissue) of the cyclic AMP response produced by subsequent re-stimulation with 1, 10 or 100 M histamine. The observed histamine-induced desensitization a...

The ability of acute and chronic Li treatment to influence agonist and depolarization-induced phosphoinositide metabolism was examined in rat cerebral cortex slices. After acute treatment (6.75 mEq/kg, 18 hr), [3H]inositol phosphates accumulating in the presence of 100 microM carbachol (muscarinic), 31 mM K+, 300 microM histamine (H1) and 300 microM 5-hydroxytryptamine (5-hydroxytryptamine2) we...
